Not sure what the spot is on your spider but it's definitely a female. It could be a small area where it may have gotten hurt and molted out of an old wound.

Before molt it was that dark spot and now it’s some white growth in it.

Could be old hemolymph from a hole or injury. I'm not sure. It's difficult to tell in the picture.
